Abstract
La traducción bacteriana es una de las dianas más importantes de los antibióticos aminoglicósidos. Estudios cinéticos y estructurales han revelado el mecanismo canónico de la traducción, así como los cambios en el mecanismo ocasionados por algunos de estos antibióticos. La fase de iniciación de la traducción es el primer punto de acción que puede tener uno de estos antibióticos. En ella, los factores de iniciación 1 (IF1), 2 (IF2) y 3 (IF3) son requeridos para garantizar el correcto ensamblaje de los complejos ribosomales de iniciación. Los antibióticos aminoglicósidos estreptomicina, espectinomicina y kasugamicina se unen a la subunidad ribosomal 30S, lo cual podría alterar la estructura y la correcta formación de los complejos de iniciación y, como acción prolongada, tener una consecuencia en la traducción bacteriana. Para poder distinguir los cambios estructurales se empleó la espectroscopía de fluorescencia basada en la transferencia de energía de resonancia de Förster (FRET), que es utilizada para evaluar los cambios en las distancias entre los fluoróforos donador y aceptor (par FRET) de los factores de iniciación modificados IF1D4C488 e IF3E166C540Q. Esta técnica ha sido empleada con anterioridad en estudios cinéticos de la formación de complejos intermediarios de la traducción, tanto en cinética pre estacionaria como en equilibrio químico. En la presente tesis se han evaluado los cambios de señal FRET en un complejo intermediario de la iniciación de la traducción, compuesto por la subunidad ribosomal 30S y los factores de iniciación modificados IF1D4C488 e IF3E166C540Q para poder ser usado como sensor de unión de los antibióticos aminoglicósidos estreptomicina, espectinomicina y kasugamicina a la subunidad 30S. El sensor ha sido probado en equilibrio químico y validado a nivel pre estacionario en tiempo real. El sensor mostró cambios opuestos en la señal FRET ante estreptomicina y kasugamicina, mientras que no mostró cambios ante la espectinomicina. Estos resultados son acordes a los diferentes mecanismos de acción de los antibióticos mencionados, y abre las puertas a futuros ensayos cinéticos con este par FRET como sensor.
Bacterial translation is one of the main targets of the family of aminoglycoside antibiotics. Kinetic and structural studies have revealed the canonical mechanism of translation, as well as changes in the mechanism caused by some of these antibiotics. The translation initiation phase is the first point of action that one of these antibiotics can have. In it, initiation factors 1 (IF1), 2 (IF2) and 3 (IF3) are required to guarantee the correct assembly of ribosomal initiation complexes. The aminoglycoside antibiotics streptomycin, spectinomycin and kasugamycin bind to the 30S ribosomal subunit, which could alter the structure and accurate formation of initiation complexes and, as a prolonged action, have a consequence on bacterial translation. Förster resonance energy transfer (FRET)-based fluorescence spectroscopy was used to distinguish structural changes by evaluating changes in the distances between the donor and acceptor fluorophores (FRET pair) of the modified initiation factors IF1D4C488 and IF3E166C540Q. This technique has previously been used in kinetic studies of the formation of translation intermediate complexes, both in a pre-steady state kinetics and in chemical equilibrium. In this thesis, the changes of a FRET signal in an intermediary complex of translation initiation, composed of the 30S ribosomal subunit and the modified initiation factors IF1D4C488 and IF3E166C540Q have been evaluated to be used as a sensor for the binding of the streptomycin, spectinomycin and kasugamycin antibiotics to the 30S subunit. The sensor has been tested in chemical equilibrium and validated at a pre-stationary level in real time. The sensor showed opposite changes in the FRET signal to streptomycin and kasugamycin, while it did not show changes to spectinomycin. These results are consistent with the different mechanisms of action of the mentioned antibiotics and open the doors to future kinetic tests with this FRET pair as a sensor.
